It can have spectacular results
Answers started to arrive in the 1990s. James Allison, at the Cancer Research Laboratory at the University of California, Berkeley, began work on a protein called CTLA-4 on the surface of some T-cells. By 1996 he had shown that this protein put a brake on the immune response to cancer. Blocking CTLA-4 with an antibody removed the brake; the immune system activated itself and got to work. Tumours in mice vanished when the animals were given CTLA-4-blocking antibodies. Though it was not immediately obvious, in retrospect this came to be seen as one of the reasons IL-2 never really worked: it is not possible to make a car run faster if its brakes are jammed on.
At the time oncologists were unimpressed by Dr Allison’s results. Cancer had been cured in mice many times over. And after many failed trials, immunotherapy was in exile—banished to the small corners of the big oncology meetings. But in 1999 Tasuku Honjo of the University of Kyoto, in Japan, showed that the gene for a protein called PD-1 also seemed to tamp down the immune system. When this gene was switched off in mice, some developed autoimmune diseases—a sign of an immune system in overdrive. In collaboration with Arlene Sharpe and Gordon Freeman at Harvard, Dr Honjo showed that some cancer cells had a second protein called PD-L1 on their surfaces which, by interacting with the PD-1 on T-cells, protected the cancer from them. Dr Honjo remembers approaching many companies with the finding, but “none wanted to invest.”
Despite a general wariness on the part of pharma companies, though, a trickle of development on therapies aimed at CTLA-4 and PD-1 did begin. Then, in 2010, Bristol-Myers Squibb released results from a trial of an anti-CTLA-4 antibody, Yervoy (ipilimumab), in malignant melanoma. Compared with the state of the art, they were fantastic. It was the first drug shown to change survival in this devastating disease, raising the median to ten months. Some survived much longer.

This Loan Agreement (this "Agreement") is entered into on April 8, 2014 by and between:
(1) Zhejiang Tmall Technology Co., Ltd., a limited liability company incorporated under the
Company Law of the People's Republic of China, with its registered address at 2nd Floor
No. 168, Wuchang Avenue, Wuchang Street, Yuhang District, Hangzhou (the "Lender"); and
(2) Simon Xie (谢世煌), with his ID number of (the "Borrower").
WHEREAS:
A. The Borrower intends to invest in Hangzhou Yunxi Investment Partnership Enterprise
(Limited Partnership) (杭州云溪投资合伙企业（有限合伙）) (the "Limited Partnership")
and becomes a limited partner thereof;
B. The Lender intends to provide financing to the Borrower, so that the Borrower may fulfill the
investment to the Limited Partnership as set out above;
NOW, THEREFORE, through amicable negotiation, the Parties hereby agree as follows:
1. Loan Commitment
Pursuant to the terms and conditions set forth herein, the Lender agrees to grant the Borrower a loan
with an aggregate amount of up to RMB 6,537,000,000 (the "Loan Commitment").
2. Grant of Loan and Purpose
2.1 The Parties agree that, during a period from the date of this Agreement to December 31, 2014
(the "Drawdown Period"), the Borrower has the right, by sending a written notice to the
Lender, to request the Lender to make payment of any part of the Loan Commitment which
has not been drawn down, in whole or in part, in no later than 10 working days, provided that
the pledge agreement as contemplated in Clause 6(2) below has been duly executed and come
into effect. The day on which the Lender makes available to the Borrower relevant loan under
a drawdown request by the Borrower is the "Actual Payment Date" in respect of such loan. In
case of any part of the Loan Commitment, which has not been drawn down by the Borrower
upon the expiration of the Drawdown Period, the Lender may cancel such part of the Loan
Commitment, and is no longer obliged to make such part of the loan under this Agreement
available to the Borrower.
2.2 Unless otherwise required by the Borrower in the drawdown request, the Lender shall make
payments of the loan to the Borrower's account as follows:

Concessione del Finanziamento

In caso di alcune parti dell’Impegno all’Erogazione, che non sono state prelevate dal Beneficiario al momento della scadenza del Periodo di Prelievo, il Finanziatore potrebbe annullare tale parte dell’Impegno all’Erogazione, e non è più tenuto a rendere tale parte del prestito in base al presente Accordo a disposizione del Beneficiario.
2.2 Salvo diverse richieste da parte del Beneficiario nella richiesta di prestito, il Finanziatore dovrebbe effettuare i pagamenti del finanziamento sul conto del Beneficiario come segue:
