English term

templated

English Science Biology (-tech,-chem,micro-) Biochemistry
Dear Colleagues,

I'd be very grateful if you could shed some light on the meaning of the word 'templated' in the following context:

'The method most frequently used to make capped RNAs in vitro is to transcribe a DNA template with either a bacterial RNA polymerase or a bacteriophage RNA polymerase in the presence of all four ribonucleoside triphosphates and a cap dinucleotide such as m7G(5′)ppp(5′)G. The polymerase initiates transcription with a nucleophilic attack by the 3′-OH of the Guo moiety in m7GpppG on the α-phosphate of the next templated nucleoside triphosphate, resulting in the initial product m7GpppGpN. The alternative, GTP-initiated product pppGpN is suppressed by setting the ratio of m7GpppG to GTP between 5 and 10 in the transcription reaction mixture.'

Full version of the document is available at http://www.google.com/patents/US20060252115.

Thank you for your assistance.

There is lots of excellent detailed material online about the mechanism of action of RNA polymerase.
Peer comment(s):

agree Polangmar : OK, this should do: http://tinyurl.com/lqj366v , http://tinyurl.com/ls9qoqa - Kowalska is the patent's co-author, Grudzień is often cited in it (but another, more detailed rewording would be welcome).
