Für den Nachweis von ASPV-spezifischen Antikörpern
wurde ein weiteres Fragment des GenoTubes zunächst in
200 μl des ELISA-Verdünnungspuffers (ID Screen® African
Swine Fever Indirect ELISA, ID Vet, Grabels, Frankreich)
überführt und dann über Nacht inkubiert. Alle weiteren
Schritte sowie die Verdünnung der originalen Serumproben
erfolgten nach Angaben des Herstellers (Protokoll für
Filterpapierstanzen). Die Ergebnisse wiesen auch hier
eine enge Korrelation zwischen den Antikörperleveln der
Tupfer- und der korrespondierenden Serumprobe auf. Ein
Verdünnungseffekt der GenoTube Proben tritt in diesem
Fall nicht in Erscheinung, da die Serumproben ebenfalls
verdünnt werden. Die Sensitivität betrug 93,1 Prozent, mit
vier falsch negativen Ergebnissen, während die Spezifität bei
100 Prozent lag.
Lateral Flow Device (LFD)
Um die Eignung einer komplett dezentralen Diagnostik zu
prüfen, wurden die kürzlich kommerzialisierten Antikörper
Lateral-Flow-Devices der Firma Ingenasa (INGEZIM PPA
CROM) erprobt. Im Detail wurde ein weiteres GenoTube
Fragment in 200 μl Verdünnungspuffer inkubiert. Im
Anschluss wurden 50 μl der so verdünnten Probe im LFD
auf ASPV-spezifische Antikörper getestet. Diese Ergebnisse
wurden mit den Resultaten des GenoTube ELISA verglichen:
Die Sensitivität der LFDs lag bei 96,2 Prozent und die
Spezifität bei 93,8 Prozent
Translation - English Antibody detection
For the detection of ASPV-specific antibodies, an additional fragment of the GenoTubes was first transferred into 200 µl of the ELISA-dilution buffer (ID Screen® African Swine Fever Indirect ELISA, ID Vet, Grabels, Frankreich) and then incubated overnight. All of the additional steps, as well as the dilution of the original sample were performed in accordance with the manufacturer’s instructions (protocol for filter paper punches). The results also showed a close correlation between the antibody levels for the swabs and the corresponding serum samples. In this case, no dilution effect of the GenoTube samples was apparent, as the serum samples were also diluted. The sensitivity was 93.1 percent, with four false negative results, while the specificity was 100 percent.
Lateral Flow Device (LFD)
In order to test the suitability of a completely decentralized diagnostics test, the recently commercialized Lateral Flow Devices antibodies from Ingenasa (INGEZIM PPA CROM) were tested. In detail, a further GenoTube genasa (INGEZIM PPA CROM) were tested. In detail, a further GenoTube fragment was incubated in 200 µl of dilution buffer. This was followed by placing 50 µl of the diluted samples into LFD and testing for ASPV-specific antibodies. These results were compared with the results from the GenoTube ELISA test: the sensitivity of the LFDs was 96.2 percent and the specificity was 93.8 percent.
