Chinese to English: Regulation of Phase Separation by Post-translational Modification of Histone General field: Science Detailed field: Biology (-tech,-chem,micro-)
(1) Transformation: A total of 50μl BL21 competent (a special physiological state of cells in which cells can take up DNA molecules from the surrounding environment, and do not decompose easily due to internal restriction endonuclease) Escherichia coli cells were added to the DNA plasmid of BRD4 protein, heated in a 42℃ water bath for 90s, and then kept on ice for 2 mins. Subsequently, the cells were added to an 800µl LB liquid medium (non-resistant) and cultured on a shaker at 37°C for 1h. Then, a solid medium containing kanamycin was prepared to inoculate the bacteria which was cultured overnight.
(2) Selecting the colony: After the colony had been cultured in the medium, the bacteria were transferred into a new liquid medium. To prevent hybrid bacterium, we picked a single point to ensure that the bacteria cultivated were what we needed. For this, 2ml LB culture solution containing 50ug/ml kanamycin was added to a 15ml corning tube, and the tweezers were heated over a flame and then used to pick a single point with a pipette tip within the solid medium. The corning tube was kept slightly flat, and the tweezers inserted in the entire pipette tip to complete the transfer.
(3) Culture rotation: after being cultured overnight, the bacteria cultured in the medium contained the specific plasmids due to the appropriate environment. The bacteria were then transferred to 500ml fresh medium with the same concentration as kanamycin at a ratio of 1:30 for continuous culture at 37°C, until the OD value reached 0.8 (for measuring the growth of flora with a cuvette and an UV spectrophotometer, similar to checking the density of flora); subsequently, 1mM IPTG was added to culture at 16°C for 16h.
(4) Protein detection: The shaken bacteria were taken out for sample retention. In each small tube 1ml of medium was added, and then centrifuged at 12,000 rpm for 5 mins to separate the bacteria from the medium. Then, the supernatant medium was removed with a pipette, leaving only the flora. A loading buffer containing bromophenol blue and SDS was added and then boiled at 95°C for 30 mins (to make the protein into a stretched state) and then taken out. The electrophoresis equipment was assembled, and the running buffer was prepared (containing Tris, SDS and glycine). The buffer was poured into the inner layer and then overflowed to the outer layer until it was flush with the inner layer. The boiled sample was taken out with a pipette and then poured through the teeth of a comb. The power was set at 70V for 15 mins processing, and then 160V for 1h processing. Gel staining was then conducted with Coomassie Blue to get the staining results to verify the expression of BRD4.
(5) Separation and purification: The bacteria of another group (3) were centrifuged for 30 mins and then the thalli were collected. Ultrasonic lysis was performed on the bacteria (energy 605J; 40°C; Pulse-ON Time: 10s; Pulse-OFF Time: 15s; Process Time: 10 mins). After that, the resulting supernatant was centrifuged again to separate the cell membrane and other contents. Finally, the interest protein was separated and purified with a nickel column. Buffer A was prepared from 50mM Tris and 500mM NaCl. After equilibration and sample loading of the nickel column, buffer A was added to the nickel column which was flushed with 50mM imidazole to ensure the equilibrium. This step was repeated. Then, the nickel column was flushed with buffer A and 100mM imidazole. This step was also repeated. Subsequently, the nickel column was flushed with buffer A and 250mM imidazole, and this step was repeated three times. The effluents were collected in separate tubes. The resulting protein was subject to another electrophoresis test to obtain the molecular weight to confirm the acquisition of the purified BRD4 protein.
