Source text - English 6 PERFORM A MEASUREMENT
• Place the test sample with the coated side up inside the clamp. Do not exceed the panel thickness as stated
in the specifications and on the machine. Testing on thicker panels can damage the drive shaft and cause
deformations that could render the device unusable.
• Close the camp to hold the panel in place. Do not use force when clamping, otherwise when removing the
panel the required force to release the panel can be to high for the operator.
• Turn on the micrometer.
• Gently turn the handle to raise the indenter. To get an indentation speed of about 0.2mm/s requires about
half a revolution per second.
• The test is carried out either to a predetermined indentation depth at where the panel is evaluated or
defects in the coating or to the depth where the first defects in the coating form.
• After the results have been determined turn the indenter back to below the zero position.
• Release the sample clamp and remove the sample.
7 INTERPRETATION OF THE RESULTS
Cracks and defects in the coating will form when the stresses exceed the max flexibility of the coating. Generally the forming of such cracks is preceded by a small discoloration of the coating. The visibility of the cracks is very depending on the type and color of the coating, just as the visual capability’s of the operator. An optional maximum magnification of 10x may be used when evaluating the samples. Please refer to the appropriate standard for further information and possibility’s regarding magnification and illumination.

• Vložte testovací panel do raznice vrstvou pokrytou nátěrovou hmotou směrem nahoru. Tloušťka testovacího panelu nesmí
přesáhnout maximální hodnotu uvedenou ve vlastnostech přístroje a na přístroji samotném. Použití panelů s větší tloušťkou může
poničit hnací hřídel a způsobit deformace, a přístroj se tak může stát nepoužitelným.
• Uzavřete komoru raznice tak, aby testovací panel pevně držel na svém místě. Při utahování stahovacího kroužku nepoužívejte
přílišnou sílu, mohli byste tak znemožnit následné uvolnění.
• Zapněte mikrometr.
• Zlehka otočte klikou pro vysunutí razníku. Pro rychlost hloubení kolem 0,2 mm/s je třeba půl otáčky klikou za sekundu.
• Posunujte razník, dokud nedosáhne předem stanovené hloubky pro vyhodnocení deformace panelu, nebo do projevení prvních
známek praskání nátěru.
• Po odečtení výsledků měření vraťte razník do původní pozice (pod úroveň okraje raznice).
• Uvolněte stahovací kroužek a vyjměte testovací panel.
7 VYHODNOCENÍ VÝSLEDKU MĚŘENÍ
Praskliny se v nátěru projeví ve chvíli, kdy tlak raznice přesáhne maximální tažnost/pružnost nátěrové hmoty. Prasklinám v nátěru
většinou předchází drobná změna barvy nátěrové hmoty. Viditelnost prasklin se z velké části odvíjí od druhu a barvy nátěrové hmoty, stejně jako od zrakových schopností obsluhy. Při vyhodnocování výsledků testování je možno využít až desetinásobného zvětšení použitím lupy s osvětlením. Pro bližší informace a možnosti zvětšení a osvětlení náhledu testovaného vzorku prosím vyhledejte příslušnou normu.

Source text - English What is DNA and where is it stored?
The nucleus is a membrane bound organelle that contains the genetic information in the form of chromatin, highly folded ribbon-like complexes of deoxyribonucleic acid (DNA) and a class of proteins called histones.
When a cell divides, chromatin fibers are very highly folded, and become visible in the light microscope as chromosomes. During interphase (between divisions), chromatin is more extended, a form used for expression genetic information.
The DNA of chromatin is wrapped around a complex of histones making what can appear in the electron microscope as "beads on a string" or nucleosomes. Changes in folding between chromatin and the mitotic chromosomes is controlled by the packing of the nucleosome complexes.
DNA or deoxyribonucleic acid is a large molecule structured from chains of repeating units of the sugar deoxyribose and phosphate linked to four different bases abbreviated A, T, G, and C. We will later show how the simple structure of DNA contains the information for specifying the proteins that allow life. The process of mitosis is designed to insure that exact copies of the DNA in chromosomes are passed on to daughter cells.

Po rozdělení buňky jsou tato mnohonásobně přeložená chromatinová vlákna viditelná a ve světelném mikroskopu se jeví jako chromozomy. Během interfáze (dělení DNA) je chromatinové vlákno více roztaženo, což je vlastně forma vyjádření genetické informace.
DNA je v chromatinu obalená kolem histonů a dohromady tvoří nukleozom, který se pod elektronovým mikroskopem podobá "navlečeným korálkům". Na smrštění a roztažení chromatinového vlákna má vliv způsob sbalení nukleozomového komplexu.
DNA neboli deoxyribonukleová kyselina je makromolekula, která se skládá z řetězců opakujících prvků cukru deoxyribózy a fosfátové skupiny spojené s jednou ze čtyř bází označených A, T, G a C. Později si ukážeme, jak je v uspořádání DNA uložena informace nezbytná pro určení životně důležitých proteinů.
